Rukhadze M, Bezarashvili G, Alexishvili M, Fischer N, Sebiskveradze M, Okujava N, Kirvalidze L, Mathematical description of the dose-dependent pharmacokinetics of Phenobarbital, Online Journal of Veterinary Research 11 (1) : 75 -88, 2007 Pharmacokinetic plasma phenobarbital concentration curves in the rabbit after different oral doses were derived mathematically. Humped ascending branches appeared in the curves at high doses. Absorption and elimination kinetics were determined using mathematical successive substitution with correlation and regression analysis versus rate constants of absorption and elimination, as well as a kinetic order of elimination. Increased dosing significantly affected the pharmacokinetics of phenobarbital. The findings suggest a step-wise absorption of phenobarbital into blood.
